Collateral sensitivity of drug resistant cells to tamoxifen is dependent on P-gp expression- Panel (A) shows a Western blot analysis demonstrating the expression of P-gp in total protein extracts from wild-type Chinese hamster ovary cells (AuxB1), drug resistant selected cells (CHORC5), P-gp-knockout drug sensitive cells (AuxB1ΔP−gp), and P-gp-knockout CHORC5 (CHORC5ΔP−gp−A1, and CHORC5ΔP−gp−A3) cells, probed with P-gp-directed monoclonal antibody (C219 mAb) and anti-α-tubulin. α-tubulin expression is shown as a loading control. Panel (B) shows the proliferation of the above cell lines (AuxB1, CHORC5, AuxB1ΔP−gp, CHORC5ΔP−gp−A1, and CHORC5ΔP−gp−A3) without or with increasing molar concentrations of colchicine (0–10 µM) or tamoxifen (0–50 µM).
