Regulation of cell viability and apoptosis by leptin is mediated by CCN5. a-b BC Cells were grown for 24 h in a 96- well plate under serum free condition. Cells were then treated with leptin (3.125 nM) or in combination of hrCCN5 (10.29 nM) in serum free media for 24 and 48 h. Cell viability was measured using crystal violet staining assay. Values on the bargraph represent the cell viability in treated and untreated groups. The data represents mean ± SEM of three independent experiments. c MCF-7 cells were serum deprived for 24 h and then treated with leptin (3.125 nM) in the presence or absence of hrCCN5 (10.29 nM) for 48 h under serum deprived conditions. Apoptotic cell death was determined using cell-death detection ELISA kit (detailed explanation in text). Values on the bargraph represent the apoptosis in treated and untreated groups.
