HDAC inhibitor sensitivity assay. The number of viable A375 cells (upper panel) was reduced by Butyrate 2 mM (by 3 %), TSA 100 nM (by 10 %), Valproic acid 1 mM (by 15 %) and Vorinostat 3 μM (by 10 %) compared with the untreated control (set at 100 %). HT-144 cells (lower panel) were more sensitive to HDACi and its viability was reduced by Butyrate 2 mM (by 13 %), TSA 100 nM (by 17 %), Valproic acid 1 mM (by 22 %) and Vorinostat 3 μM (by 25 %). Cell number was determined after 24 h of treatment by the MTT proliferation assay (Sigma). Shown data correspond to representative experiments carried out in triplicate and repeated twice.
