FnIII-1c desensitizes NCI-H460 cells to TRAIL-induced apoptosis. NCI-H460 cells were pre-treated with 10 μM FnIII-1c, FnIII-10n, FnIII-13, or PBS for 1 h, then stimulated with 50 ng/ml TRAIL for an additional 2.5 h. a Cell lysates were analyzed by western blotting for expression of cleaved caspase 8 and GAPDH. b Densitometric quantification of the data shown in (a). Corresponding densitometry for cleaved caspase 8 presented as a change in protein expression relative to TRAIL-treated cells. Each bar represents the mean ± SE of three independent experiments. A one-way ANOVA with Tukey post-hoc analysis was used to determine statistical significance (*p < 0.05). c Apoptotic Cells were visualized by TUNEL assay and stained with Hoescht 33342 dye to denote nuclei. Bar = 50 μm. d The percentage of TUNEL positive cells was determined by Image J analysis. Each bar represents the mean ± SE of five independent experiments. A one-way Anova with Tukey post-hoc analysis was used to determine statistical significance (*, p < 0.05)
