In combination assays, NSCLC cell lines were seeded in 96-well plates at a density of 10⁵/well with culture medium for 24 h. The cell lines were treated with FU (10 μM) alone or FU plus gefitinib (1 μM)/mithramycin A (50 nM) for another 24 h at 37 °C in a 5 % CO₂ atmosphere. After that, WST-1 was added to each well and incubated for 2 h at 37 °C before measuring absorbance at 490 nm with a Multiskan JX Spectrum instrument.
