Silencing of human microRNA miR-21 expression by siRNAs. Quantitative real time RT-PCR analysis of miR-21 expression in SiHa cells transfected with pSIMIR21 plasmid. Total RNA and cDNA synthesis were obtained from 1 × 105 SiHa cells (HPV16+) per well in a six-well plate containing DMEM at 37 °C with 5 % CO2 after 48 h transfection with pSIMIR21 plasmid (0, 0.5, 1, 3 and 5 μg). Relative expression by real-time RT-qPCR analysis of miR-21 was calculated using the 2-∆∆Ct method and was normalized by miR-21/RNU6 ratio relative expression units. The Ct values were analyzed with pSilencer 1.0-U6 empty vector transfection and pSIMIR21 plasmid and values are presented as mean ± SD. The P values <0.01 are indicated with asterisks
