TSG101 depletion has little impact on the signaling pathways in PMA-stimulated HT1080 cells. Cells transfected with control (con) or TSG101 (TSG#1) siRNA were incubated in fresh serum-free medium containing the indicated reagents. a–b. Cells were incubated with 50 μM PD98059, 10 μM SB203580, 50 μM SP600125, or 50 μM LY294002 in fresh serum-free medium containing or not 200 nM PMA for 7 h. MMPs in conditioned media from PMA-treated (a) and non-treated (b) cells were measured using gelatin zymography. c. Cells were incubated in fresh serum-free medium containing 200 nM PMA for the indicated periods of time. Cell lysates were analyzed by western blot using the indicated antibodies.
