A: Geranyl-geranyl transferase I (GGTase I) was quantified using an in vitro test based on [3H]-prenyl pyrophosphate (0.5 μM, 8–10 Ci/mmol) incorporation into a mutant form of H-ras with a geranyl-geranylation CAXX box. The level of prenylation is expressed as a percentage of maximum incorporation of [3H]-prenyl, as determined by allowing the uninhibited reaction to go to completion.
