S37F mutant β-catenin activity in medulloblastoma DAOY cells. Cotransfection of the TOPLASH Wnt reporter and FLAG-tagged S37F mutant β-catenin into DAOY cells results in 41-fold higher luciferase activity than transfection of TOPFLASH alone. Similar transfections of wild-type β-catenin result in 3.8-fold higher luciferase activity. No luciferase activity was detected in cells transfected with pBluescript (Negative Control).
