Effects of transient silence of Tβ10 on cell migration in KKU-M214 cell line. (A) KKU-M214 cells were treated with 50 pM Tβ10 siRNA or scramble siRNA as a control for 48, 72 and 96 h, respectively. Tβ10 mRNA and protein levels were determined by real time RT-PCR and immunohistochemistry, respectively. (B) Migration and (C) invasion assays were carried out in KKU-M214 cells using a modified Boyden chamber assay. Cells were pre-treated with Tβ10 siRNA or scramble siRNA for 48 h, and placed on Transwell to determine cell migration (15 h) and on Transwell with pre-coated Matrigel (18 h) to determine cell invasion.
