Suppression of SNCG in LNCaP Cells inhibits cellular proliferation and induces cell cycle arrest at G1. (A) Top: RT-PCR analysis was used to evaluate SNCG mRNA expression levels in androgen-dependent (LNCaP) and androgen-independent human advanced prostate cancer lines (DU145, PC3 and LNCaP-AI). Bottom: Western blot analysis revealed SNCG protein expression levels in the relevant cell lines. (B) Top: RT-PCR analysis was used to evaluate SNCG mRNA expression in LNCaP cells after transient transfection with various siRNA constructs. Bottom: Western blot analysis showed oligo-166 inhibited SNCG expression in LNCaP cells. (C) CCK-8 assay were performed for evaluation of the LNCaP cellular proliferation. (D) Flow cytometry assay demonstrated that inhibition SNCG expression in LNCaP cell induced cell cycle arrest at G1.
