Allocryptopine fails to induce apoptosis at concentrations below 250 μM. Allocryptopine-induced apoptosis was quantified by flow cytometric analysis using light scatter characteristics (white bars), measurement of mitochondrial membrane potential Δψm (grey bars) and the activation of caspase-3 (black bars) upon treatment of Jurkat Vector cells for 24 h with medium or 0, 50, 100, 150 and 250 μM allocryptopine. Data show means ± SD (n ≥ 3) (left panel). Fluorescence microscopy upon combined staining with Hoechst33342 and propidium iodide staining of cells treated with 250 μM allocryptopine for 24 h showed increased numbers of cells with apoptotic morphology upon treatment (right panel).
