DNA damage assessed by histone γH2AX and 53BP1 foci counts per nucleus in control or drug-treated and/or irradiated M059K (a) and M059J (b) cells. Cells were treated with PI-103 3 h prior to irradiation with 2 Gy, fixed at different time points (30 min, 2 h, 4 h and 6 h) after irradiation and immunostained for γH2AX and 53BP1. DNA damage was assessed by the mean number of γH2AX (green) and 53BP1 (magenta) foci per nucleus in non-irradiated and irradiated (2 Gy) cells, either in control or in cells treated with PI-103 fixed 30 min, 2 h, 4 h and 6 h post-irradiation. For each time point at least 100 nuclei were evaluated using the ImageJ plugin FocAn [28]
