IGFBP-3 mediates the effect of VD3 on cell growth in LNCaP cells. a Overexpression of IGFBP-3 in LNCaP cells. LNCaP cells were infected with lentivirus containing the IGFBP-3 gene, and its overexpression was confirmed by quantitative reverse transcription–polymerase chain reaction. b Suppression of cell growth by IGFBP-3. IGFBP-3–overexpressed cells were cultured as they were and control cells were cultured with DHT/VD3 for 3 days, and then the cell number was measured. c Knockdown of IGFBP-3 in LNCaP cells. LNCaP cells were infected with lentivirus containing shRNA for Igfbp-3, and IGFBP-3 knockdown was confirmed by quantitative reverse transcription–polymerase chain reaction and western blotting. The ratio indicates the density of IGFBP-3 band normalized by corresponding βActin band. d The IGFBP-3 knockdown cells were treated with DHT and/or VD3 for 3 days, and then the cell number was measured.
