Gating strategy for analysis of expression of IFN-γ or granzyme B by CD8⁺ subpopulations. Blood leukocytes were immunostained for CD4, CD8 CD3, IFN-γ or granzyme B. CD8⁺CD4⁻ cells were then identified as cells that expressed higher levels of CD8 plus CD3 (CD8^hiCD3⁺), low levels of CD8 plus CD3 (CD8^dimCD3⁺) or low levels of CD8 but not CD3 (CD8^dimCD3⁻). Each of these subpopulation was then gated on and analyzed for their expression of IFN-g or granzyme B
