Impairment of HR repair by miR-7-5p in Dox-resistant SCLC cells. a Reporter construct used for the analysis of HR repair. After the introduction of DSBs by I-SceI digestion, HR repair was measured by GFP gene reconstitution in cells containing a single copy of a chromosomally integrated HR repair substrate (DR-GFP). b SCLC cells were cotransfected with I-SceI and pDRGFR plasmids, and then the percentage of GFP-positive cells was measured by FACS. c HR repair activity was impeded by miR-7-5p in SCLC cells. H446AR and H446 cells were transfected with miR-7-5p mimic or inhibitor, respectively, and the reporter construct was transiently transfected 24 h later, followed by the analysis of the GFP-positive cells by FACS.
