SSd triggered apoptotic factor release in HSC-T6 cells. The mitochondrial (a) and cytosolic (b) fractions were isolated following the treatment of HSC-T6 cells with 1 μM SSd. The purities of mitochondrial and cytosolic fraction were validated with anti-COX3 and anti-GAPDH antibodies by Western blotting. The expression levels of Apaf-1, Cyt c, EndoG and AIF were detected by Western blotting with specific antibodies in mitochondrial (c) and cytosolic (d) fractions. (e) HSC-T6 cells were grown in 24-well chamber cover glasses; treated with 1 μM SSd for 60 min; stained with MitoTracker® Deep Red FM (100 nM) for 30 min; fixed with 4 % cold paraformaldehyde, and incubated with specific primary antibodies and FITC-conjugated secondary antibody. DAPI was adopted as a nuclear counterstain. The fluorescence signals were analyzed by confocal laser scanning microscopy
