Apoptosis was evaluated by ELISA using a Cell Death Detection ELISA^Plus Kit (Roche Applied Science). To evaluate the effect of gefitinib and mithramycin A on cell apoptosis, we treated cells with gefitinib (1 μM) and mithramycin A (50 nM) for 24 h. Extracted cell lysates were evaluated by ELISA. In wild-type cells (H1299, H1437) there was little effect of the inhibitors on apoptosis. By contrast, apoptosis increased in EGFR-mutated cell lines treated with gefitinib, except for H1975 with the T790M mutation. Mithramycin A increased apoptosis in all EGFR-mutated cell lines.
