From: Epigenetic-based combinatorial resveratrol and pterostilbene alters DNA damage response by affecting SIRT1 and DNMT enzyme expression, including SIRT1-dependent γ-H2AX and telomerase regulation in triple-negative breast cancer

Fig. 8

Combination treatments altered epigenetic enzyme expression and their activity in HCC1806 breast cancer cells. a Resveratrol and pterostilbene after 72 h of treatment affected DNMTs (DNMT1, DNMT3A, DNMT3B) mRNA expression. With the combination of resveratrol and pterostilbene all three DNMTs were significantly down-regulated. Resveratrol at 15 μM also resulted in a decrease in DNMT3B mRNA expression after 72 h. GAPDH was used as an internal control. b Overall DNMT enzyme activity analysis, using 20 μg of nuclear extract. After 72 h, the entire treated group resulted in a significant down-regulation of enzyme activity in comparison to DMSO control. There was no significant difference in overall enzyme activity within the treatment group itself. Values are representative of three independent experiments and represented as percent of control ± SE; *P <0.05, **P <0.01
