Clonogenic survival to oxidative stress. 1.0 × 106 cells were grown for 24 h in 37°C and 5% CO2 incubators. Following this incubation, the cells were washed with 1× PBS and treated with H2O2 in PBS for 30 min. The cells were washed twice with 1× PBS and incubated at 37°C, 5% CO2 in their respective media for 2 h. Serial dilutions of the cells were made in media and the dilutions were plated. The cells were then incubated at 37°C, 5% CO2 for 20 days. The cells were washed twice with 1× PBS and then stained with 0.02% crystal violet. The surviving colonies were scored using MCID computer software. The symbols are, diamonds for AG11134, squares for MCF-7 and triangles for HCC1937. The experiments were performed in triplicates and the error bars represent standard error of mean.
