Figure 1

(A) Effect of exogenous H2O2 on MRP1 expression. EAC/Dox cells were incubated overnight in presence of 50 μM H2O2. Surface MRP1 expression was studied by flow cytometry and confocal microscopy. Representative data of 5 independent experiments is presented. (B) ROS generation by EAC/Dox cells ex vivo 24 h following in vivo CuNG treatment. CuNG treatment given after 7 days following inoculation with EAC/Dox cells. Results are compared with ex vivo ROS generation by EAC cells derived from EAC/S and EAC/Dox bearing mice. EAC/Dox cells following in vivo CuNG treatment and EAC/S cells generated significantly higher levels of ROS compared to EAC/Dox cells from untreated animals (*, P < 0.001 and **, P < 0.005). Results are presented as mean ± S.D. of 3 independent experiments. Each experiment was done with 5 animals in each experimental group. (C) Confocal microscopy of the effect of CuNG on MRP1 expression in EAC/Dox cells. EAC/Dox cells expressed higher levels of MRP1 than EAC/S cells and CuNG treatment reduced MRP1 expression. Results presented are representative of 3 independent experiments.
