In vitro angiostatin generation by tPA and D-penicillamine. Murine serum was incubated with tPA, D-penicillamine, or a combination of the two. Reaction products were subjected to Western blot analysis employing an anti-mouse plasminogen antibody. In control samples (a) or samples incubated with D-penicillamine (c), no conversion of plasminogen could be demonstrated. Incubation of serum with tPA resulted in the formation of plasmin and a small amount of angiostatin (b), whereas substantial amounts of angiostatin were observed after incubating serum with both tPA and D-penicillamine (d).
