V158411 reduces cell viability and induces caspase-3/7 dependent apoptosis. A. V158411 inhibits the viability of triple-negative breast and ovarian carcinoma cells. Cells were exposed to 5 or 10-times the GI50 of V158411 for 72 hours and the cell viability determined using trypan blue staining. Values are the average of ≥4 determinations ± SD. B. Caspase-3/7 dependent apoptosis is induced in triple-negative breast and ovarian carcinoma cell lines in vitro. Cells were exposed to 10-times the GI50 of V158411 for 24 or 48 hours and caspase activity determined using a homogenous caspase-3/7 activation kit. Values are the average of 3 replicates ± SD. C. Caspase-3/7 activation was confirmed by immunoblotting in SKOV-3 cells treated for 48 hours with the indicated concentrations of V158411. FL, full length; CL, cleaved.
