LCL85 treatment decreases xIAP protein levels in mouse tumor cells. A & B. Colon 26 (A) and 4 T1 (B) cells were cultured in the absence or presence of LCL85 (10 μM) and FasL (Colon 26 = 250 ng/ml; 4 T1 = 24 ng/ml) as indicated for 48 h. Cells were then stained with Annexin V and PI and analyzed by flow cytometry. The apoptotic cell death is indicated at the top right corner of each plot. C: Quantification of apoptosis. % FasL-induced cell death is calculated by the formula: % Annexin V⁺ and PI⁺ cells in the presence of FasL - % Annexin V⁺ and PI⁺ cells in the absence of FasL. D.
