In vivo local, passive anaphylaxis in human FcεRIα transgenic mice. Mice were injected intradermically on the back with PBS alone, 1μg of PSA IgE or a NS IgE. After 4 hours, mice were injected intravenously with either A) 25 μg anti-human κ, B) 50 μg of the HSA-PSA peptide (containing multiple epitopes for AR47.47) conjugate, or C) 50 μg PSA in 1% Evans blue in PBS. Animals were euthanized 10 minutes later. A local hypersensitivity/anaphylactic response (acute inflammatory reaction) caused by the degranulation of FcεRI-expressing cells in the skin is visualized by leakage of the dye into the skin due to vasodilation of local blood vessels.
