Analysis of HGF-induced c-met activity in cell lines. (A) WT, NEO4 and S3DN cells were stimulated with 100 ng/mL HGF for 10 and 60 minutes. Lysates were resolved on a 4-20% SDS polyacrylamide gel, transferred to nitrocellulose, and probed sequentially with antibodies for p-Stat3, total Stat3, p-c-met, total c-met, and β-actin. (B) Bar graph of western blot quantification. Values indicate the scanned signals normalized to the β-actin signal. (-) and (+) correspond to without and with HGF treatment, respectively. (C) WT & DN2 cells were stimulated with 100 ng/mL HGF for 30 minutes and probed with a p-c-met antibody, followed by an Alexa-546 tagged secondary antibody. Panels photographed at 600×.
