Influence of pan-caspase inhibitor zVAD-fmk on growth inhibition induced by C-DIMs. MDA-MB-231 [A] and MDA-MB-453 [B] cells were treated for 48 h with 10 μM DIM-C-pPhCF3 (CF3), DIM-C-pPhtBu (tBu) or DIM-C-pPhC6H5 (C6H5) and with or without 20 μM zVAD-fmk and then counted as described in the Materials and Methods. Results of all proliferation studies are presented as means ± SE for at least three separate determinations for each treatment group. Significantly (p < 0.05) decreased growth (compared to DMSO) in cells treated with C-DIMs alone is indicated (*). FACS analysis of MDA-MB-231 [C] and MDA-MB-453 [D] cells co-stained with Annexin V and PI. Cells were treated for 48 h or 24 h with 10 μM MG132. Floating and adherent cells were incubated with Annexin V and PI and analyzed by flow cytometry as described in the Materials and Methods. Results of all proliferation studies are presented as means ± SE for at least three separate determinations for each treatment group. Significance (p < 0.05) compared to DMSO is indicated (*).
